چکیده

Introduction Parkinson’s disease (PD) is characterized by loss of dopaminergic neurons in the substantia nigra (SN), but its etiology remains poorly understood (1). In vivo H MRS may provide a noninvasive alternative to culture, animal model and post-mortem studies that are more commonly utilized to understand the pathogenesis of PD. However, due to its small size, the MRS investigation of the SN in the human brain is difficult (2). Previous studies primarily employed 6-8ml voxels which implicate substantial partial volume effects. In addition, they were performed at 1.5 Tesla and therefore only provided data on NAA, creatine, choline and their respective ratios (2). The aim of the current study was to determine the feasibility of expanding the information content of H MR spectra from smaller volumes at 4 Tesla to neurotransmitters, such as GABA and glutamate, and the antioxidant glutathione.

Proton MRS of the unilateral substantia nigra in the human brain at 4 tesla: detection of high GABA concentrations.

Parkinson's disease (PD) is characterized by loss of dopaminergic neurons in the substantia nigra (SN), the cause of which is unknown. Characterization of early SN pathology could prove beneficial in the treatment and diagnosis of PD. The present study shows that with the use of short-echo (5 ms) Stimulated-Echo Acquisition Mode (STEAM) spectroscopy and LCModel, a neurochemical profile consisti...

Elevated Pontine and Putamenal GABA Levels in Mild-Moderate Parkinson Disease Detected by 7 Tesla Proton MRS

BACKGROUND Parkinson disease (PD) is characterized by the degeneration of nigrostriatal dopaminergic neurons. However, postmortem evidence indicates that the pathology of lower brainstem regions, such as the pons and medulla, precedes nigral involvement. Consistently, pontomedullary damage was implicated by structural and PET imaging in early PD. Neurochemical correlates of this early pathologi...

Increased GABA levels in medial prefrontal cortex of young adults with narcolepsy.

STUDY OBJECTIVES To explore absolute concentrations of brain metabolites including gamma amino-butyric acid (GABA) in the medial prefrontal cortex and basal ganglia of young adults with narcolepsy. DESIGN Proton magnetic resonance (MR) spectroscopy centered on the medial prefrontal cortex and the basal ganglia was acquired. The absolute concentrations of brain metabolites including GABA and g...

Ultra-high field MRS of rodents

Rats and mice may serve as subjects for modeling various pathological conditions, which can be studied by localized MR spectroscopy. In contrast to MR imaging, which relies on properties of water related to its interaction with biomacromolecules, spectroscopy is capable of providing information on concentrations of chemical entities present in a living tissue. Measurements at high magnetic fiel...

MR detection of secondary changes remote from ischemia: preliminary observations after occlusion of the middle cerebral artery in rats.

PURPOSE To determine whether secondary MR changes occur in the thalamus or the substantia nigra after middle cerebral artery (MCA) occlusion in rats. METHODS Sprague-Dawley rats were subjected to MCA occlusion. At varying intervals, proton density-, T1-, and T2-weighted images were obtained with a 4.7-T superconductive MR unit. RESULTS T2-weighted images revealed an area of high signal inte...
